چکیده

2 Definitions  Neurofilaments: Nf are proteins which are exclusively expressed in neurons and their adjacent axons. Nf are particularly abundant in the axon, where they are key building blocks of the axonal cytoskeleton. The complex protein chemistry of Nf is briefly described.  Biomarker: A biomarker is a characteristic that is objectively measured and evaluated as an indicator of normal biologic processes, pathogenic processes, or pharmacological responses to therapeutic intervention.  Surrogate endpoint : defines a biomarker that is intended to serve as a substitute of a clinically meaningful endpoint and is expected to predict the effect of a therapeutic intervention or the evolution of disease.

Neurofilament phosphoforms: surrogate markers for axonal injury, degeneration and loss.

This review on the role of neurofilaments as surrogate markers for axonal degeneration in neurological diseases provides a brief background to protein synthesis, assembly, function and degeneration. Methodological techniques for quantification are described and a protein nomenclature is proposed. The relevance for recognising anti-neurofilament autoantibodies is noted. Pathological implications...

A TOXIC NEUROFILAMENTOUS AXONOPATHIES: ACCUMULATION OF NEUROFILAMENTS AND AXONAL DEGENERATION Suggested running headline: Neurofilamentous axonopathies

A number of neurotoxic chemicals induce accumulation of neurofilaments in axonal swellings that appear at varying distances from the cell body. This pathology is associated with axonal degeneration of different degrees. The clinical manifestation is most commonly that of a mixed motor–sensory peripheral axonopathy with a disto-proximal pattern of progression, as in cases of chronic exposure to ...

Axonal degeneration in paraplegin-deficient mice is associated with abnormal mitochondria and impairment of axonal transport.

In several neurodegenerative diseases, axonal degeneration occurs before neuronal death and contributes significantly to patients' disability. Hereditary spastic paraplegia (HSP) is a genetically heterogeneous condition characterized by selective degeneration of axons of the corticospinal tracts and fasciculus gracilis. HSP may therefore be considered an exemplary disease to study the local pro...

Neurofilaments are nonessential to the pathogenesis of toxicant-induced axonal degeneration.

Axonal neurofilament (NF) accumulations occur before development of symptoms and before other pathological changes among idiopathic neurodegenerative diseases and toxic neuropathies, suggesting a cause-effect relationship. The dependence of symptoms and axonal degeneration on neurofilament accumulation has been tested here in a transgenic mouse model (Eyer and Peterson, 1994) lacking axonal NFs...

Slow axonal transport in acrylamide neuropathy: different abnormalities produced by single-dose and continuous administration.

Alterations in axonal caliber and neurofilament content have been associated with altered neurofilament transport in several models of neurofibrillary degeneration. Acrylamide intoxication provides a prototype of distal axonal degeneration, the most frequent pattern of axonal pathology in human and experimental neurotoxic injury. Neurofibrillary changes are a variable and often minor aspect of ...
